Acute toxicity, phenol content, antioxidant and postprandial anti-diabetic activity of Echinops spinosus extracts

Echinops spinosus, belonging to Asteraceae family, is used in folk medicine as an abortifacient and diuretic for blood circulation, diabetes, stomach pain, indigestion spasmolytic problems. The objective of this work the study acute toxicity, content phenolic compounds (polyphenols, flavonoids tannins), antioxidant activity (DPPH, ABTS, FRAP, H2O2 xanthine oxidase) antidiabetic (?-amylase, ?- glucosidase lipase) vitro ex-vivo by studying starch tolerance test. phytochemical assay showed that ethanolic extract richest polyphenols, tannins with 77.01 mg GEA/g extract; 544.33 RE/g extract, 32.20 EC/g respectively. better compared aqueous (IC50=13±0.25 µg/mL; IC50=75.11±0.34 TE/g IC50=51.1±1.2 AAE/g IC50=28.2±2.87 µg/mL 16.83 ± 0.72 µg/mL) DPPH, oxidase. Extracts E. spinosus have shown a remarkable inhibitory effect ?-amylase interesting ?-glucosidase lipase. also lowered sugar levels 0.96 0.93g/L, respectively, after 90 minutes starch-loaded rats. Acute toxicity results indicate extracts are non-toxic LD50 greater than 2 g/kg female Swiss mice. Therefore, anti-diabetic may be at origin bioactive contained plant spinosus. However, vivo studies on mechanism action needed against oxidative stress associated diabetes.
